Wir hatten bereits angesprochen, dass man Guitar Hero 5 nicht nur auf die Setliste „reduzieren“ sollte, dennoch… die Musik ist in einem Musikspiel logischerweise einer der tragenden Pfeiler. Activision Blizzard konnte sich auch für die 5. Auflage wieder zahlreiche namenhafte Bands und deren Hits sichern. Zudem verfügt auch Guitar Hero 5 über das aus dem Vorgänger bereits bekannte GH Studio, das es ermöglicht, eigene Beats, Synthie-Samples und Gitarrenriffs zu einem eigenen Stück zusammenzustellen, dieses dann abzumischen und auf GH Tunes hochzuladen oder Inhalte anderer Hobbykomponisten herunterzuladen.
Zusätzlich gibt es mittlerweile zahlreiche Tracks und Alben als herunterladbare Inhalte im Online-Musikladen, der in regelmäßigen Abständen mit neuen Hits gefüttert wird. Zu guter letzt ist es auch möglich, die Tracks aus Guitar Hero World Tour oder Guitar Hero Greatest Hits zu importieren. Das kürzlich erschienene Guitar Hero Metallica wird aber leider ausgesperrt.
